The peptide Selank protected against ethanol-induced memory and attention deficits in rats during alcohol withdrawal by normalizing BDNF levels in the hippocampus and prefrontal cortex.
p<0.01 memory protectionSelank significantly prevented ethanol-induced memory and attention disturbances during alcohol withdrawal in rats
What the researchers found
Selank at 0.3 mg/kg/day for 7 days intraperitoneally produced a cognitive-stimulating effect in 9-month-old control rats (p<0.05) and prevented ethanol-induced memory and attention disturbances during alcohol withdrawal (p<0.01). In ex vivo brain tissue analysis, Selank prevented the ethanol-induced increase in BDNF content in both the hippocampus and frontal cortex (p<0.05). The protective effect on memory was assessed using the object recognition test, which measures both recognition memory and attention.
Why it matters
Alcohol-related cognitive impairment is a major public health problem with few effective treatments. This study suggests Selank could protect against alcohol-induced brain damage through a specific mechanism — normalizing BDNF levels. The fact that Selank also improved cognition in non-alcohol-exposed older rats suggests broader neuroprotective potential.
How the study worked
Outbred rats received 10% ethanol as their sole fluid source for 30 weeks to model chronic alcohol exposure. Selank was administered intraperitoneally at 0.3 mg/kg/day for 7 days. Memory and attention were assessed using the object recognition test. BDNF content was measured ex vivo in the hippocampus and frontal cortex. Both ethanol-exposed and control (non-exposed) 9-month-old rats were tested.
What this study cannot tell us
This was an animal study in outbred rats, and results may not translate directly to humans. The sample size was not specified in the abstract. The 7-day treatment period is very short, and it's unclear if protection persists after Selank discontinuation. The ethanol-only fluid model is extreme and may not represent typical human drinking patterns. Selank is not FDA-approved and is primarily used in Russia.
How to read the evidence
This is a controlled animal study with statistical analysis and ex vivo mechanistic confirmation through BDNF measurement. However, it's limited to a single rat study without human validation.
When this study was published
Published in 2019, this study contributes to the ongoing characterization of Selank's neuroprotective properties. Selank remains primarily available in Russia and is not approved by the FDA or EMA.
The bigger picture
Selank is a Russian-developed peptide anxiolytic based on the endogenous immune peptide tuftsin. This study adds to evidence that Selank has cognitive-enhancing properties beyond its anti-anxiety effects, specifically through BDNF-mediated neuroprotection. It fits into growing interest in peptide-based neuroprotective agents for addiction-related brain damage.
Questions still open
- Would Selank provide similar cognitive protection in humans with alcohol use disorder during withdrawal?
- Why does chronic ethanol exposure increase BDNF in the hippocampus, and why does preventing this increase improve cognition?
- Could Selank be used as a neuroprotective adjunct during alcohol detoxification protocols?
Common questions
How does Selank protect against alcohol-related memory loss?
Does Selank improve memory in people who don't drink alcohol?
